Protective human hybridoma antibody to tetanus toxin.

Postimmunization human B lymphocytes and mouse myeloma cells were fused to produce interspecies hybridomas secreting human antibody of predefined specificity with an initial frequency comparable to intraspecies fusion. After 13 mo in culture, one clone continued to secrete high titers of human IgG antitetanus toxin antibody. This antibody binds to the B fragment of tetanus toxin and protects mi...

Toxin-neutralizing effect of antibody against subtilisin-digested tetanus toxin.

A form of systemic tetanus with atypical symptoms was observed in mice injected in the left thigh with a mixture of tetanus toxin and antibody produced in guinea pigs against a fragment of toxin obtained from a subtilisin digest of the crystallized toxin. The mice did not show typical symptoms of the local tetanus such as convulsions or spastic paralysis of the injected limb.

Diversity of the Antibody Response to Tetanus Toxoid: Comparison of Hybridoma Library to Phage Display Library

Monoclonal antibodies are important tools in research and since the 1990s have been an important therapeutic class targeting a wide variety of diseases. Earlier methods of mAb production relied exclusively on the lengthy process of making hybridomas. The advent of phage display technology introduced an alternative approach for mAb production. A potential concern with this approach is its comple...

Characteristics of human antibody to diphtheria toxin.

Human antibody to diphtheria toxin was analyzed with an ammonium sulfate radioimmunoassay. In 15 immune sera with titers of between 0.03 and 25 units/ml, the avidity was similar to that of U.S. standard antitoxin. The proportion of the post-immunization anti-diphtheria antibody specific for fragment A was variable, but in the 10 sera tested constituted a substantial fraction of the antibody. Ap...

Identification of human and rodent thymic epithelium using tetanus toxin and monoclonal antibody A2B5.

Using a monoclonal antibody (A2B5), which binds to GQ ganglioside, and tetanus toxin, which binds to GD and GT gangliosides, distinct regions of human and rodent thymic epithelial cells have been identified. The lymphoid elements of the thymus do not bind A2B5 or tetanus toxin. The A2B5 and tetanus toxin-binding cells form a network of thymic epithelial cells throughout the thymic subcapsular c...
